WELCOME TO THE 45th ANNUAL CONFERENCE OF THE AMERICAN COLLEGE OF VETERINARY OPHTHALMOLOGISTS By Dr. Ralph Hamor, Program Chair

It is with pleasure that I welcome each of you to Fort Worth, Texas for the 45th Annual ACVO Conference at the lovely Omni Fort Worth Hotel. We hope that this conference provides a wide variety of educational and social opportunities for our attendees, both at the meeting and in the greater Dallas/ Fort Worth area. There should be many wonderful opportunities waiting for you to discover. Our William Magrane Memorial Speaker this year will be Dr. Robert Noecker, who is the director of glaucoma at Ophthalmic Consultants of Connecticut in Fairfield, CT. His talk is titled: “Advances in Glaucoma, Cataract and Anterior Segment Surgery. Dr. Noecker is a leading ophthalmologist in the fields of medical, laser and surgical therapy of glaucoma and cataracts. He is currently an Assistant Clinical Professor at Yale School of Medicine. He has lectured all over the world and his presentation should be a treat. We continue to have a large number of submissions for abstracts and posters…almost 130 this year. The Planning Committee hopes that the presentations will provide attendees access to stimulating new ophthalmic information. As in the past few years, we will continue grouping our scientific presentations into subject areas. This will allow us to provide the best opportunity to meet continuing education requirements for some of our members. It also allows attendees to more readily compare presentations in similar subject areas. We will have a break out session for equine ophthalmology on Friday that is sponsored by the IEOC. Our poster sessions on Thursday and Friday will also be grouped by subject areas and are sponsored by Ocu-GLO. The pre-conference Practice Management Course will be held on Wednesday afternoon and features Leslie Mamalis MBA, MSIT, CVA. Ms. Mamalis is the owner of Summit Veterinary Advisors, a leading veterinary management and financial consulting firm. Her team works exclusively with veterinarians and is known for expertise in specialty practice valuation. There are two post-conference courses on Sunday: the Genetics Symposium and the Comparative Ocular Surface Disease Workshop. The Genetics Symposium will have internationally known speakers helping to guide participants on how to utilize known advances in genetic discovery to provide appropriate counseling to our clients. The Comparative Ocular Surface Disease Workshop will allow participants to focus on presentations that are focused on ocular surface disease. The keynote speaker will be Dr. Bernardo Yanez-Soto from the University of California-Davis. The Resident’s Workshop features Dr. Tammy Miller Michau and Dr. James Morrisey. They will be sharing their expertise in their presentation titled: “Veterinary Ophthalmology in Exotics”. Once again this year, current or former residents have submitted publications to compete for an award in the areas of Basic Science, Clinical Research or a Case Report/ Review Article. We continue to be impressed with the quality of these publications from our residents. We will again be providing a General Practitioners course on Saturday. Thank you to all of the speakers who are participating. We hope that this course will continue to generate interest in ophthalmology as well as be informative to the local veterinarians in the greater Dallas/Fort Worth area. We are happy to once again have Dr. Amber Labelle organize the 4th annual ACVO Juried Photography competition. The ACVO Vision for Animals Foundation will have a cocktail party and auction on Thursday evening sponsored by Prescription Center. Please plan to send in your photos for the competition and participate in the VAF auction. As always, we will have a Welcome Reception on Wednesday evening and a Social Dinner on Friday evening. The Fort Worth area provides numerous opportunities to explore including the Stockyard National Historic District, the World Renowned Cultural District, Sundance Square and the Fort Worth Zoo. These events and opportunities should provide ample opportunity to enjoy the area and/or network with your colleagues. I would like to thank all of the sponsors and vendors that are here for both their generosity and their time. Please take time to thank them and to visit their booths. I have to thank everyone who has been involved in the planning of this meeting. Until you have been involved in the planning of a meeting of this size and scope, it is impossible to understand all that is involved. I am thankful for the generous assistance of my planning team members, Drs. Elizabeth Giuliano and Jane Cho. I could not have done any of this without their guidance, service and support. All three of us want to express our extreme gratitude for the expertise and assistance of Stacee Daniel and her amazing staff. Words cannot in any way convey all of their effort to make this meeting a success for our members. It continues to be an honor to serve as a member of the Board of Regents as I have been so blessed by this organization and its members. I ask that each of you continue to be, or consider becoming, an active member of this organization. We need each member’s input as we navigate the years ahead.

COMPARISON OF NEPAFENAC, DICLOFENAC AND PRANOPROFEN FOR REDUCING INTRAOCULAR INFLAMMATION IN DOGS. (R. Kato, N. Takiyama, A. Sugiyama, T. Harada) Laboratory of Veterinary Internal Medicine, Department of Veterinary Medicine, College of Bioresource Sciences, Nihon University, 1. Purpose To compare effects of topically administered nepafenac, diclofenac and pranoprofen for controlling aqueocentesis-induced uveitis in dogs. Methods. Dogs were allotted to 4 groups (n=10 per group). Dogs in each groups received a topical 0.1% nepafenac (Alcon Japan, Tokyo, Japan), 0.1% diclofenac (Wakamoto Co., Tokyo, Japan), pranoprofen (Senju Pharmaceutical Co., Osaka, Japan) or artificial tear (Senju Pharmaceutical Co., Osaka, Japan) served as control. One hour after an applied eye drop, dogs were sedated and an initial aqueocentesis of 0.3 mL was performed. One hour later, a second aqueocentesis was performed. All aqueous samples were kept in −80°C until assayed for PGE2 concentrations with ELISA kit (R&D Systems, Prostaglandin E2 Parameter Assay kit®, Minneapolis, MT, USA). Results. Mean PGE2 concentration of first samples was 53.5 pg/mL. Mean PGE2 concentrations of second samples were 49.5 ± 63.7 pg/mL (nepafenac), 24.5 ± 77.6 pg/mL (diclofenac), 277.6 ± 268.1 pg/mL (pranoprofen), and 2125.7 ± 262.5 pg/mL (artificial tear). Mean PGE2 concentrations of second samples of nepafenac, diclofenac, and pranoprofen groups were significantly lower than that of control group. Nepafenac and diclofenac resulted in a nearly complete inhibition of PGE2 synthesis (P < 0.01 vs. control) and was a greater effect than pranoprofen (P < 0.05). Conclusions. Nepafenac and diclofenac were more effective than pranoprofen for controlling the production of PGE2 in dogs with experimentally induced uveitis. None.

Canine corneas were collected and used to generate primary canine corneal fibroblasts (CCFs) by growing cultures in minimal essential medium supplemented with 10% fetal bovine serum. Canine corneal myofibroblasts (CCMs), used as a model of canine corneal fibrosis, were produced by growing CCF cultures in serum-free medium containing transforming growth factor β1 (1 ng/ml). Trypan blue viability assays determined the optimal pirfenidone dose for this in vitro model while phase contrast microscopy, and TUNEL assays were used to evaluate potential cytotoxicity. Scratch and MTT assays evaluated the effect of pirfenidone on cellular migration and proliferation. Real-time PCR, immunoblot analysis, and immunocytochemistry were employed to determine the efficacy of pirfenidone to inhibit CCM formation in vitro. Results. Treatment with 200 μg/ml pirfenidone significantly decreased aSMA expression when compared to the TGFβ1 control group (P < 0.01). Pirfenidone treatment ≤ 200 μg/ml did not affect CCF phenotype or cellular viability and did not result in significant cytotoxicity. Conclusions. Pirfenidone safely and effectively inhibits TGFβ1-induced myofibroblast proliferation in the canine cornea in vitro. In vivo studies are warranted. OCULAR AND SYSTEMIC SAFETY OF TOPICAL SUBEROYLANILIDE HYDROXAMIC ACID (SAHA) IN DOGS (KM Gronkiewicz 1, EA Giuliano 1, A Sharma 1, RR Mohan 1,2,3) College of Veterinary Medicine, University of Missouri;1 Harry S Truman VA Hospital, Columbia, Missouri; 2 Mason Eye Institute, University of Missouri.3 Purpose. To evaluate the local and systemic effects of topical Vorinostat® (suberoylanilide hydroxamic acid, SAHA), a drug with significant anti-fibrotic potential, in canine corneas in vivo. Methods. Six purpose-bred beagles underwent a safety trial, receiving either topical A) vehicle (DMSO diluted in sterile PBS, 2 μL/mL), B) low dose (25 μM) SAHA or C) high dose (50 μM) SAHA. Daily ophthalmic examinations were performed. Ocular health was graded according to the modified McDonald-Shadduck (mMS) scoring system. Complete blood counts, serum biochemistries and urinalyses were collected on days 0 and 5. Following a washout period of 5 days, Latin Square Allocation was used to re-assign dogs to new treatment groups, and the safety study was repeated until each dog received topical treatments A, B and C at least once. mMS scores and laboratory tests were analyzed with repeated measures one-way ANOVA and Holm-Sidak test post-hoc, with p
