Tertiary structure of a DNA aptamer developed by Base Pair Biotechnologies

Prior to use aptamers need to be resuspended and folded into their tertiary structure for optimal binding Handling and Storage Store lyophilized aptamers at -20 ºC for ideal long term stability. Aptamer Resuspension Protocol Briefly centrifuge the aptamer storage tube to ensure the dried aptamer pellet is at the bottom of the tube. Resuspend the aptamers pellet in TE buffer (10 mM Tris HCl, 0.1 mM EDTA, pH 7.5) or non-DPEC treated, nuclease free water to achieve100uM concentration (using volumes indicated in the Product Information Sheet that accompanies each order). Incubate at room temperature for 30 minutes. Briefly vortex and quickly spin the sample down (~10,000×g for 1 second). Create aliquots and store –20ºC. Aptamer Folding Instruction Before Use Prior to use, dilute the aptamer to its working concentration in Folding Buffer:1 mM MgCl 2, 1X PBS (137 mM NaCl, 2.7 mM Potassium Chloride, 8 mM Sodium Phosphate dibasic, 2 mM Potassium Phosphate monobasic), pH 7.4.

Heat the aptamer solution to 85 — 95 ºC for 5 minutes. Allow the solution to cool to room temperature (~15 minutes). Aptamers are now ready for use and will not need to be folded again.
