BIOTECHNOLOGY Imaging techniques have been developed into powerful tools over the last decade. Confocal microscopy offers "optical sectionning" and 3-D reconstruction without the need for mechanical sectionning; the applications and limitations of this technique as applied to the fungal system are reviewed by Czymmek, Whallon & Klomparens (1994). Another technique for imaging at the molecular level is scanning tunneling microscopy. Muller et al. GENETICS, MOLECULAR BIOLOGY & EVOLUTION One of many interesting papers in this issue deals with aspects of transcription. Shea, Toyn & Johnston (1994) isolate and clone the DBF3 gene of Saccharomyces cerevisiae and confirm its function in the S phase as a cell cycle regulated protein kinase. But they also discover that its sequence is identical to PRP8, a gene coding for the protein component of the U5 small ribonucleoprotein, associated with the spliceosome during transcription. Kollar et ai. (1995) examine the linkage between chitin and 13(1- > 3)-glucan in S. cerevisiae by enzymatic digestion and NMR spectroscopy while Shibata et ai. (1995), using similiar techniques as well as agglutination inhibition assays, analyse the different branched side chains of Candida mannan. Lu et al. (1995) use kre mutants and immunoprecipitation to demonstrate that "a-agglutinin is covalently associated with j31,6-glucan" in the cell wall of S. cerevisiae and that this linkage is "dependent on prior addition of a cpr anchor to a-agglutinin N.
