COMBINED EFFECTS OF TESTOSTERONE PROPIONATE AND Leptadenia hastata PERS. (DECNE) AQUEOUS EXTRACTS ON IMMATURE CASTRATED MALE RATS Bayala Balé, UFR/SVT, University of Ouagadougou, BURKINA FASO
Karlsruhe, 28-29 November 2012, Germany, Alexander Von Humboldt Foundation Meeting INTRODUCTION Leptadenia hastata (Pers.) Decne. is a perennial liana species of Asclepediacea family and it is widely used in African tradi9onal medicine. L. hastata have been reported to be rich in steroidal glycosides, cardenolides, flavonoids, triterpenes and polyoxypregnane deriva9ves. In the north of Burkina Faso, where livestock is very important, the breeders claimed the loss of fer9lity of their animals aEer the consump9on of L. hastata leaves. The purpose of the present study was: first, to determine the compe99on between L. hastata extracts and testosterone propionate (TP) as androgen reference in vivo; and second, to evaluate the an9-‐androgenic ac9vity of L. hastata extracts. MATERIAL AND METHODS 150 g of the leaves of L. hastata were collected, washed, dried, macerated and lyophilized. Three weeks old Wistar male rats were castrated and randomized into 2 treatment groups. Each group was divided into 4 subgroups of 6 animals. The four subgroups of the first group received dis9lled water (as control), 100, 200 and 400 mg/kg of L. hastata aqueous extracts respec9vely. For the second group, each subgroup received in combina9on 0.4 mg/kg of TP and the same amounts of L. hastata aqueous extracts. TP was administered subcutaneously and L. hastata extracts by oral gavage during 10 consecu9ve days. 24 h aEer the last administra9on, the rats were killed and androgen-‐ Bayala et al. 2927 dependent sex glands weight were measured. Fructose and prosta9c acid phosphatase (PAP) were also measured in prostate and seminal vesicle.
