MATERIALS AND METHODS: Study Subjects: The screening study group included 255 generalized vitligo patients and 377 unrelated controls, all of Indian subcontinent (Pakistan, India, Sri Lanka, Bangladesh) derivation (IP). The replication study group included 685 unrelated generalized vitiligo patients and 774 ethnically-matched non-vitiligo controls from Gujarat state, India. The diagnosis of generalized vitiligo was based on standard diagnostic criteria (Taieb and Picardo, 2009). Controls had no other known autoimmune diseases. This study was approved by the Combined Institutional Review Board (COMIRB) of the University of ColoradoDenver, Aurora, CO USA; by the Ethics Committee of King Edward Medical University, Lahore, Pakistan; and by the Institutional Ethical Committee for Human Research (IECHR) of The Maharaja Sayajirao University of Baroda, Gujarat, India, and was conducted in accord with the Declaration of Helsinki Principles. Written informed consent was obtained from all study subjects.

Genotyping Genomic DNA was prepared from peripheral-blood specimens by standard methods or from saliva specimens with use of a DNA self-collection kit using the manufacturer’s instructions (Oragene, DNA Genotek). DNA concentrations were assayed by ultraviolet A260 spectrophotometry (Nanodrop, Thermo Scientific). For the large-scale study genotyping was performed by the University of Colorado Denver Genomics and Microarray Core laboratory using the Immunochip

(Illumina), and quality control filtering of genotype data was performed using Illumina GenomeStudio version 3 and PLINK version 1.07 (http://pngu.mgh.harvard.edu/purcell/plink/). SNPs were excluded on the basis of call rate
