Introduction

Dear Colleagues, Metallomics Technology Conference is an international forum for scientific discussion focused on understanding the relationship and connections between metals and their binding species inducing amino acids, peptides, proteins and nucleic acids. The first of conferences was held in Brno from 14 to 16 June 2015 and brought a lot of new knowledge gathered in the conference proceedings, which is available in printed and electronic form. The aim of this multidisciplinary conference is to bridge the gaps between the specialists and fields of science as diverse as inorganic chemists, biochemists and clinicians. There is a need for sensitive and reliable analytical methods which would help us to determine what bioactive metal-binding compounds are found in an organism, as well as for trace analysis methods in complex biological matrices to follow the bioactive compounds and their metabolites in the human body. Moreover, we need to understand how disorders and illnesses can influence these. It is evident that enormous progress has been made in metallomics over the last decade. Recent challenges for scientists in this field are to develop analytical technologies that would allow us to understand our personal metabolism, in connection with specific biomarkers, with the overall aim to personalize medicine as generalized recommendations and to discover how to influence metal metabolism for treatment of civilization diseases. The second of conferences was held as a videoconference from 25 to 26 August 2015. Its main topic was metallomics and analytical methods. Present conference proceedings contains not only presented papers but also three press releases, which inform about main activities of the Metallomic Scientific Network, which included, among other events, also two mentioned conferences.

Summary Essential oils are important natural compounds in anti-therapeutic treatment. They represent complex systems showing sometimes very complicated effects in different micro-organisms. Interested to study the antibacterial properties of these natural extracts, we focused our attention on the inhibition effect of the steam distillation extracts obtained from Saturea Montana, Myrtus communis, Laurus nobilis, Juniperus communis, Salvia officinalis and Thymus vulgaris on two pathogenic bacteria colonies such as Staphylococcus aureus and Escherichia coli. In each case, two different temperatures (20 °C; 37 °C) and oil volumes (5 μL; 20 μL) were considered within a time range of 24 hours. At 37 °C, all the employed essential oils presented antibacterial effects and these effects were intensified with the increasing of the essential oil volumes used. The essential oils of Saturea Montana, Laurus nobilis and Thymus vulgaris revealed a higher inhibition rate on Staphylococcus aureus than the others, while the essential oils of Saturea montana, Laurus nobilis, Myrtus communis and Thymus vulgaris were more efficient on the inhibition of Escherichia coli. Although a reasonable inhibiting effect of the essential oils was observed even at 20 °C, a temperature decrease from 37 °C to 20 °C diminished the inhibition effectiveness of some essential oils. Due to the observed antibacterial properties, all these essential oils are recommended to be used as additives in medicinal and cosmetic products.

1. Introduction More than 1340 plants are known to be potential sources of antimicrobial compounds, but few have been studied [1]. Several other works have examined the effect of compounds isolated from essential oils extracted from plants on fungi and bacteria [2]. Nowadays, most of the studies are concentrated in the mixture of different quantities of essential oils and inhibition of various bacteria and mycoses. The main goal of these analyses is related with the use of essential oil in shampoos, creams, perfumes and other cosmetic productions. Nowadays the scientists are studying deeply the antimicrobial effect of essential oils and their active components. Essential oils with their main components, including alcohols as well, can act as strong antibacterial agents. This action is due their stability, especially based on pH point of view, whereas the antibacterial effect of phenols becomes vulnerable from a decrease level of pH. Some scientists give arguments that this inhibitory mechanism is related to the bacterial cell wall composition and structure. Many oils and extracts from medicinal plants have been surveyed for their bactericide action against a series of bacterial colonies. Essential oils are found to contribute in the therapeutic treatment of tumors. Some oils have shown to 28

have conservation properties on foods, pharmaceutics, whereas some other are used successfully in aromatherapy. Often the chemical compounds used for therapeutic treatments or as preservatives mainly in food chemistry are associated with long term collateral effects in humans or animals. Therefore, the natural antimicrobial compounds use, is important and much more acceptable not only in food conservation but also in the control of humans and microbial diseases. Our study gives a further contribution in this broad field. It is mainly concentrated in the inhibitory effect of six essential oils against two bacterial colonies Staphylococcus aureus and Escherichia coli. The gram positive bacteria, such as Staphylococcus aureus are primarily responsible for post-operative wound infections, toxic shock syndrome, endocarditis, osteomyelitis and food poisoning [3]. Escherichia coli, which is the one of the gram negative bacteria, present in human intestines and causes urinary tract infection, coleocystitis or septicemia [4]. These two potentially pathogenic microorganisms were used to evaluate antimicrobial activity of essential oils, such as: Saturea montana, Myrtus communis, Laurus nobilis, Juniperus communis, Salvia officinalis and Thymus vulgaris.

2. Materials and methods The necessary materials for this study are: alcohol lamps, microscope glass, microscope, lighter, thermostat, fridge, Petri dishes, cotton, rinsing solution, water.

2.1. Plant materials and the essential oils In the present study we used the essential oils of Saturea montana, Myrtus communis, Laurus nobilis, Juniperus communis, Salvia officinalis and Thymus vulgaris, because these plants are well known for their medicinal properties and are easily available in our country. They were by provided “XHERDO” company (½ L essential oil for each medicinal plant), which obtains them by water steam distillation.

2.2. Micro-organisms The micro-organisms used for this purpose were obtained by two different patients. Staphylococcus aureus is obtained from a patient pustule using a sterile tampon. The bacteria are cultivated in blood agar ground, which is suitable for the bacteria colony cultivation and growth. The incubation temperature was 37 0C and the incubation period was 24 hours. Escherichia coli colonies are drawn from patient feces through a sterile tampon and cultivated blood agar ground, which is suitable for the bacteria colony cultivation and growth. The incubation temperature was 37 0C and the incubation period was 24 hours. The blood agar ground, adaptable for bacteria cultivation consists of distillated water, common nourishing agar, universal peptone, meat extract, NaCl, ram blood and pH = 7.4 [5].

2.3. Inhibitory activity identification The identification method of S. aureus is realized by the positive catalase prove, which is positive only for S. aureus. Other Staphyloccocus colonies do not give a positive reaction, even if golden Staphyloccocus colonies are easily distinguished by their round, smooth and lightly form. Coagulate prove is realized in a microscope glass where are thrown two – three 29

dots physiologic solution and suspended the colonies. When the catalase is positive (when the effervescence is appeared) means that the S. aureus is present. The E. coli colonies have the same view and therefore there is not any special criterion for their selection. That’s why we chose 10 colonies from the grounds where they are cultivated (blood agar ground). These colonies are emulsified with physiologic solution and checked for an evidently agglutination in the dense suspension formed. The distinct agglutination shows the E. coli presence. The E. coli distinctions can be also done easily from the microscope [6]. The colonies of S. aureus and E. coli are treated with different quantity of essential oils in different temperature for 24 hour to identify the inhibitory effect of six essential oils in variable conditions. After the bacterial colonies (S. aureus and E. coli) are developed, the respective grounds are divided in three parts. Over each part is poured 5 µL / 20 µL essential oil. Micro organisms’ incubation is realized in two different temperatures, 37 °C and 20 °C. For this purpose six measures are done: 1, 3, 5, 8 and 24 hours after the essential oil is poured.

3. Results and discussion The first measurements are realized using a small volume of essential oil (5 µL) for each medicinal plant in a temperature 37 0C. The results of these experiments are shown in the chart below (chart 1) which presents the percentage of bacterial inhibition (Staphylococcus aureus) versus the monitoring time.

Chart 1: Evolution of the inhibitory effect of six essential oils on Staphylococcus aureus colonies when using a volume of 5 µL and a temperature of 37 °C. Chart 2: Evolution of the inhibitory effect of six essential oils on Staphylococcus aureus colonies when using a volume of 20 µL and a temperature of 37 °C.

All the essential oils taken into consideration (chart 1) show inhibitory effect toward Staphylococcus aureus colonies. For the essential oils of T. vulgaris, L. nobilis, M. communis and S. montana the inhibition effect cease after the period of contact time of 8 hours. While the inhibition effect continues, although at lower efficiency, until 24 hours for J. communis and S. officinalis. A significant antibacterial activity is observed for the essential oils of S. montana, M. communis, L. nobilis and S. officinalis. For the essential oils of S. montana 30

and S. officinalis this antibacterial effect can be attributed mainly to the presence of phenolic components. Meanwhile, other antibacterial compounds such as eucalyptol, terpineol and carvacol present in the essential oil of M. communis and Cineol- 1,8 in L. nobilis essential oil, display also a salient antibacterial effect at T = 37 °C. The next experiments performed, took into consideration higher volumes of essential oils and the same temperature for the six employed essential oils. The difference between the inhibitory effects for the two employed volumes (5 µL and 20 µL respectively) of the essential oils is obvious when comparing chart 1 and chart 2. These differences show that the specific antibacterial effect of the oils under consideration depends mainly on the volume of the essential oil used and not significantly on the temperature in which the experiments occurs. A pronounced antibacterial activity is shown by essential oils of T. vulgaris, S. montana, and S. officinalis which can be a dedicated to the presence of cineol- 1, 8, fenolic components, timol and carvacol in these oils. Insuppressible in our measurements is the salient increase of the inhibitory effect toward the Staphylococcus aureus colonies when using higher volumes of T. vulgaris essential oil in comparison to the other essential oils (steeper slope, chart 2). The temperature of the developing bacterial ground is another crucial parameter which strongly influences the bacterial inhibition effect parallel to the essential oil volume used. Our next investigation considered the decrease of developing ground temperature in the condition of constant oil volumes used. Our accurate observations related to this investigation revealed no decrease of the inhibitory effect for S. officinalis and T. vulgaris oils versus the temperature decrease. A slight diminishing of the inhibitory effect was observed for the essential oil of S. montana, meanwhile, the other essential oils showed a considerable dependence from this parameter. Indeed there are two factors which competitively decrease the inhibitory effect of the essential oils when decreasing the ground temperature. These are: a) a decrease of the temperature leads to the increase of the oil viscosity and therefore a decrease of its penetrability into the cell walls. b) As the inhibition reaction is strongly depended on the developing ground temperature, a decrease of temperature will lower the number of active molecules which have the energy equal or higher than the activation energy, and as a consequence will decrease the inhibitory effect. The antibacterial effect of six essential oils against S. aureus using 20 µL essential oil and a temperature of 20 0C is shown in chart 3. Due to the content of the above mentioned chemical compounds, significant antibacterial activity is observable for the essential oils of S. montana, S. officinalis and T. vulgaris. Other essential oils show almost a negligible inhibitory effect due to the temperature decrease.

31

Chart 3: Evolution of the inhibitory effect of six essential oils on Staphylococcus aureus colonies using a volume 20 µL and a temperature of 20 °C. Chart 4: Evolution of the inhibitory effect of six essential oils on E. coli colonies using a defined volume of 5 µL and a temperature of 37 °C.

The same experiments as above are realized for the E. coli colonies in order to compare the inhibitory effect of the essential oils toward it. The antibacterial progress of the selected essential oils toward E. coli colonies using a quantity of V= 5 µL and the temperature T= 37 °C is shown in the chart 4. Significant inhibitory effect toward E. coli colonies is presented by the essential oils of L. nobilis, M. communis, S. Montana, T. vulgaris. For all these oils, a decrease in the number of colonies of approx. 15 % is observed (Chart 4). S. officinalis and J. communis resulted less effective for the selected conditions. Similarly to the inhibition effect observed for Staphylococcus aureus colonies, an increase of the employed oil sample volume reveals better inhibition efficiency even for the E. coli colonies (chart 5). All these effects are strongly related to their antibacterial composition and specifically for M. communis, S. Montana and T. vulgaris this is due to the presence of the active compounds such as: eucalyptol, caryofilen, carvacol, terpineol and phenol and thymol. A less tilted inhibition slope is observed for M. communis and T. vulgaris for an incubation time of 8 hours. A longer contact of these essential oils with the bacterial species revealed a better inhibition effect, which probably might be dedicated to the diffusion velocity of the components of these oils in the E. coli cell walls. The temperature decrease influenced the inhibition effect of essential oils toward E. coli colonies. Again similarly to the Staphylococcus aureus colonies, almost no inhibitory effect was observed for the essential oils M. communis, L. nobilis toward E. coli colonies.

32

Chart 5: Evolution of the inhibitory effect of six essential oils on E. coli colonies using a volume of 20 µL and a temperature of 37 °C. Chart 6: Evolution of the inhibitory effect of six essential oils in E. coli colonies using a defined volume of 20 µL and a temperature of 20 °C.

The increase of the viscosities of these oils are associated to lower diffusion and though to low penetrability. The number of active molecules (molecules which enter the inhibition reaction) is also lowered due to decreasing their energy. This explanation can be generalized for the rest of the considered oils (chart 6). The inhibitory effect observed for some essential oils slightly affected by the temperature is probably due to the presence phenolic and alcoholic compounds in them.

4. Conclusions In this study we focused our attention on the inhibition effect of the steam distillation extracts (essential oils) obtained from Saturea Montana, Myrtus communis, Laurus nobilis, Juniperus communis, Salvia officinalis and Thymus vulgaris on two pathogenic bacteria colonies such as Staphylococcus aureus and Escherichia coli. The inhibitory effect of these oils was studied for two different temperatures (20 °C; 37 °C) and oil volumes (5 μL; 20 μL) within a time range of 24 hours. For the experimental conditions 5µL, 370C, the essential oils of T. vulgaris, L. nobilis, M. communis and S. montana ceased their inhibition effect on Staphylococcus aureus colonies after the period of contact time of 8 hours. While the inhibition effect continued, although at lower efficiency, until 24 hours for J. communis and S. officinalis. This antibacterial effect can be attributed mainly to their content of phenolic components. All the employed essential oils presented enhanced inhibitory effect on Staphylococcus aureus when the essential oil volume was increased from 5 μL to 20 μL at constant temperature (37 °C). At such conditions a salient increase of the inhibitory effect toward the Staphylococcus aureus colonies was observed for T. vulgaris essential oil in comparison to the other essential oils. A decrease of the ground temperature from 37 °C to 20 °C at constant essential oil volume (20 μL) revealed no decrease of the inhibitory effect of S. officinalis and T. vulgaris oils toward Staphylococcus aureus. A slight diminishing of the inhibitory effect was observed for the essential oil 33

of S. montana, meanwhile, the other essential oils showed a considerable dependence from this parameter. The experiments performed on E. coli colonies displayed significant inhibitory effect for the essential oils of L. nobilis, M. communis, S. Montana, T. vulgaris at 37 °C and 5 μL oil volumes used. S. officinalis and J. communis resulted less effective for these conditions. An increase of the employed oil volume at 37 °C revealed better inhibition efficiency for the E. coli colonies. The temperature decrease again influenced the inhibition effect of essential oils toward E. coli colonies similarly to the Staphylococcus aureus colonies. At 20 °C almost no inhibitory effect was observed for the essential oils M. communis, L. nobilis toward these bacteria.

Możliwości wykorzystania metod analitycznych w metalomice W dniach 25 i 26 sierpnia, naukowcy z Laboratorium Metalomiki i Nanotechnologii, Zakładu Chemii i Biochemii, Uniwersytetu Mendla w Brnie zrealizowali wideokonferencję pt Metalomika i Metody Analityczne. Podsumowanie wspólnej pracy naukowców z grupy Wyszehradzkiej w ramach Międzynarodowego Funduszu Wyszehradzkiego przedstawia książka streszczeń Konferencja Technologie w Metalomice 2015: Najnowsze Postępy i Strategie, lub w książce streszczeń Konferencja Technologie w Metalomice 2015: Metalomika i metody analityczne. Elektroniczna wersja tych książek streszczeń jest do pobrania ze stron internetowych projektu przedstawionych poniżej.   87

A videokonferencia témái a metallomikai kutatásban történt előrelépések, az ezzel a gyorsan fejlődő tudományterülettel kapcsolatos analitikai módszerek terén elért haladás, illetve a metallomika daganatterápiában való felhasználása voltak. A videokonferencia lehetőséget adott a Visegrádi csoport „Metallomic Scientific Network” projektben résztvevő tagjainak találkozására. A Metallomikai és Nanotechnológiai Laboratóriumban megtartott szemináriumok nemcsak a metallomikai alapkutatásról szóltak, hanem a metallomikai kutatások daganatterápiában való felhasználásáról, illetve a betegségek biológiai alapjainak jobb megértése céljából szövetmintákon (pl. csirkeembrió) alkalmazható tömeg-spektrometriáról is. A Debreceni Egyetem Általános Orvostudományi Kar Bőrgyógyászati Klinikán szervezett háromnapos workshop témája a metallomika bőrdaganatok kezelésében való felhasználási lehetőségei voltak. Minden projekt-tevékenység kapcsolódási pontjának tekinthetők azok az egynapos videokonferenciák, melyeknek központi témája a metallothionein fehérje szerepe volt a metallomikai kutatásban. A metallothioneint 1957-ben írták le először, és funkciója a mai napig nem teljesen tisztázott. Ugyanakkor ismert, hogy szerepet játszik a nehézfém-toxicitás elleni védelemben, ami jelenleg nagyon aktuális a környezet-szennyezés miatt. A metallomikai kutatás alkalmazhatósága az onkológiában is megvitatásra került. A Visegrádi csoport kutatóinak a Nemzetközi Visegrádi Alap projektje keretében megvalósult együttes munkájának összegzését adja a Metallomikai Technológiai Konferencia 2015: Legújabb Fejlesztések és Stratégiák absztrakt könyve, illetve a Metallomikai Technológiai Konferencia 2015: Metallomika és Analitikai Módszerek absztrakt könyve. Az absztrakt könyvek elektronikus verziója letölthető a lentebb jelzett web helyről.
