DETERMINATION OF KETAMINE BY LCMS/MS – A METHOD VALIDATION FOR FORENSIC INVESTIGATION Verma Kanak Lata, Verma Rohan, Panicker Veena, Sharma Madhulika, Sarin RK, Jaiswal AK* Forensic Science Laboratory, Government of National Capital Territory of Delhi, India LabWater Division, Merck Millipore, Bangalore, India Department of Forensic Medicine and Toxicology, All India Institute of Medical Sciences, Ansari Nagar, New Delhi- 110029,India Received 26-12-2014; Revised 23-01-2015; Accepted 21-02-2015 ABSTRACT A liquid chromatography-tandem mass spectrometry method was developed for the determination of ketamine for its determination in the aerated drinks and mineral water. Ketamine popularly known as date-rape drug abused in rave parties and cause of the crimes against the women and this method would be able to determine its residues in non-biological exhibits in cases related to drug facilitated sexual assaults. This method was developed to determine ketamine in cold drinks and mineral water and is able to quantify with precision. Extraction method used for the Samples (soft drinks, soda water and mineral water) was dilute and shoot to http://www.ujponline.com/ujpjournal/determinationofketaminebylcmsmsamethodvalidationforforensicinvestigation/

make it fast and at the same time reliably accurate. The total run time for the current analytical method is less than 5 minute. As the extraction procedure, Dilute and shoot method has also helped in increasing throughput so that more number of exhibits can be studied with short turnaround time. Calibration curve has been established from 0.5 to 50ng/ml for ketamine with correlation coecient exceeding 0.99. The limit of detection and the limit of quanti cation obtained were 0.1 and 0.5 ng/ml respectively for ketamine. We have also studied the impact of quality of pure water used for the extraction procedure for ketamine from various drinks like drinking water and aerated drinks which are generally the forms in which ketamine is prescribed to victims. We have observed the clear di럐erences in results amongst the tap water, double distilled water and ultrapure water (Milli Q water) used in extraction procedure. The results show that the proposed method is suitable for testing this drug in any sample of aerated drink and mineral water. In addition, it is simpler and faster than analysis by conventional methods such as gas chromatography-mass spectrometry, which usually require a more laborious extraction procedure. In the view of the simplicity and sensitivity the proposed method is recommendable for the determination of ketamine using high purity water in non-biological exhibits with special reference to DFSA Cases.
