ABSTRACT The mass application of the drug praziquantel (PZQ), 2-(cyclohexylcarbonyl)1,2,3,6,7,11b-hexahydro-4H-pyrazino[2,1-a]isoquinolin-4-one, to large volume seawater aquariums is a new procedure being used to control the parasitic trematode Neobenedenia melleni, but no method for detecting PZQ in seawater has been published. The animal care staff at The Living Seas Pavilion at the Walt Disney World® Resort requested that such a method be created before they endeavored to treat their six million gallon seawater aquarium with PZQ. A highperformance liquid chromatography (HPLC) method to determine PZQ levels in seawater was developed at Disney’s The Living Seas for their first large-scale treatment of PZQ in order to confirm that the target concentration was achieved and to track how long the PZQ remained in the water. The extraction technique and HPLC analysis method will be discussed as well as real life treatments.
